Abstract

Esterification reactions between L-alanine and various sugars were performed in organic solvent using lipase from Rhizomucor miehei as the biocatalyst. Furthermore, esterifications between L-alanine and glucose catalysed by the R. miehei enzyme or porcine pancreatic lipase were optimized in terms of enzyme and substrate concn., pH, buffer concn. and reaction time. Sugars used as substrates in the microbial lipase-catalysed reaction are glucose, galactose, mannose, fructose, arabinose, ribose, lactose, maltose and sucrose and conversion yields with these sugars ranged from 8 to 56%. For the microbial enzyme, the optimum conversion yield (28%) was achieved using 30% enzyme, 1mM glucose, 4mM alanine, 0.1mM acetate buffer and pH 4.0 for 72 h. The max. conversion yield for the pancreatic lipase was 18% and was achieved under the following reaction conditions: 40% enzyme; 1mM glucose; 2mM alanine; 0.1mM acetate buffer; and pH 5.0 for 72 h. Activity of the microbial lipase, but not porcine pancreatic lipase, was enhanced in the presence of buffer salts. The mono- and di-esters produced from these reactions were identified using NMR.
