Additional Information: Incidence of diabetes has been growing at an alarming pace not only in the developed countries but also in the developing countries especially in India. Type 1 diabetes mellitus (T1DM) is usually a progressive autoimmune disease, resulting from an insidious immunologic destruction of pancreatic beta cells and is the result of multiple genetic and environmental influences. It is unknown which one first starts this cascade of immune events, but immune events important in this process include destruction of beta cells of the islets of Langerhans by immune factors called cytokines which are produced by the T lymphocytes. The HLA class II genes are most strongly associated with T1DM. Further, by comparison of T1DM of human and rat genome, there were similarities and evolutionary relationship between them. Globally many research institutes/ laboratories commonly use laboratory rat (Rattus norvegicus) as animal model for development and evaluation of anti-diabetic compounds. But the complete metabolic pathway for T1DM in rat is naive. Considering this, an attempt is made to evaluate the genomic relation between human and rat genome with reference to T1DM. For this purpose, the onine Database KEGG for downloading TIDM genes from Pathway, BLAST tool for finding nucleotide sequence similarity between them and Map Viewer to visualize the BLAST hit on the genome of Rat and identify the gene corresponding to the type 1 diabetes mellitus genes, UniGene database is used to collect the information unigenes are identified for aligned genes, ScanProsite to detect the conserved domain and Cn3D to visualize the structure-based sequence alignments and SWISS-MODEL for automated comparative modeling in MHC II and PROCHECK for Ramachandran plot analysis. Comparative genome analysis between them reveals that ten genes of TIDM shows similarities with laboratory rat chromosomes i.e. MHC1, MHC11, CPE, ICA, Fas, FasL, GAD, HSP, IA-2 and TNFB. These genes are showing Genes and Unigenes in laboratory Rat Genome. By using homology modeling 3D structure for hypothetical protein LOC688090 is predicted using Swiss model. Their sequence similarity in rat gives a strong evidence of presence of the conserved domains in genomic composition and thereby chances to produce functionally similar proteins in T1DM pathway. Thus the study reads the use of laboratory rat as a suitable animal model for human T1DM, also paves the way to the computational prediction and construction of metabolic pathway for rat. Structure prediction of hypothetical protein will be much helpful in identification and designing of lead compounds for drug discovery.
