Item Type: Student Project Report
Additional Information: The synthetic polymer of polyethylene accumilation was the major problem in our daily life. These plastics not only take many decades to be decomposed in nature but also produce toxins during the process of degradation. Biodegradable plastics repeseted a solution to environmental problems generated by utilization of plastics from in the petrochemical sources, which have many undesirable properties such as resistance to degradation and durability. PHA is biodegradable, echo friendly non toxic and it degraded easily by microorganims. PHV is the most known PHA, PHB is accumilated in a large no of bacteria under the conditions of nutrient limitation and in the presence of an abundant souce of carbon. Some of the PHA producing microorganism synthesize the exopolysaccharacrides in the fermented medium. The Rizobium meliloti produced more PHA than others (Zebenhuizen, 1986). The objective of the present work study about the conditions of exetracellular polymers of Sinorhizobium MTCC 100. The PM was suplimented with CSOC and CCOC to compaire the PHA PM. The quality of polymers was characterized by GC analysis. The optimization studies were carried of sucrose as carbon source to produce biopolymers. The amount of biomass (6.92 g/l) was produce more than other concentration. The production of biomass was more in CCOC oil cake extract and minimum PHA (26.54). The pH and incubation peried also importent in the production of biopolymers by Sinorhizobium MTCC 100. The pH 8 was found to be suitable to synthesize extra cellular polymers by Sinorhizobium MTCC 100. The rhizobium microorganism are synthesize more polymers in this condition. The kinetics of polymers production by Sinorhizobium MTCC 100 was studied up to 96 hrs. From the results, the 24 hrs of fermentation resulted in production of higher amount of PHA (27.2%). The exopolysaccharide characterization was analysing by GC analysis showed multiple peaks indicating that the presence of PHB and PHV. It was concluded that agro residues can be exploited for enhanced production of biopolymers from Sinorhizobium bacteria. As all the experiments of the present study were carried out in short time. Some more studies is required for conformation.
