Additional Information: Bacillus cultures in general are designed as starter culture to meet food safety, shelf life, technological effectiveness and economical feasibility criteria. Specially selected cultures may also provide probiotic benefit, and, if properly modified, they may even be endospored with nutraceutical traits. Not all desirable probiotic chareristics were present in a single isolate, where many isolates displayed varying individual but promising capabities. The study titled “ charecterization of probiotic properties of native Bacillus species” includes analysis of the major probiotic requirements in native Bacillus isolates. The isolates were studied for their biochemical properties and found to that all the isolates were catalase positive and could survive at anaerobic conditions. Isolates 27, 40 and 50 were found to hydrolase starch and these isolates were determined to be non pathogens by lecthinase and hemolysis test. In case of physiological test, all the organisms were found to grow at 50◦C and could grow in the presence of 8% bile concentration which indicates that these isolates are capable of surviving in the gut ( ˜0.3%) which is an ideal criteria for a probiotic. It was found that all the isolates showed growth at high pH ( 8 & 10) indicating that they are alkalophilic but at low pH (pH 2 & 4) no growth was observed. When compared to B.coagulans isolate no 26, 16 showed better growth in the presence of acid (pH2) and isolates no 16, 27, 50 were tolerant to bile (0.3%), which is a major criteria of an probiotic bacteria. Isolates no 27 and 16 showed better adhering and autoaggregation when compared to B.coagulans and isolates no 50 showed similar percentage. All the isolates showed above average percent of scavenging activity which is additional property of a probiotic bacteria. Isolate no 27 showed the antimicrobial activity when compared to other isolates on all indicator organisms. It was found that non of the isolates were active against E.coli and isolates no 16 was found inactive against L.monocytogens and S.aureus. similarly isolate no 50 was found inactive against L.moncytogen. isolates no 27, 40, 50 and control organism showed positive results for BSH test, indicating that these native Bacillus strains are capable of reducing serum cholesterol which is one of the important property of the probiotic bacteria. From the above analysis it was found that the isolates no 16, 27, 50 can be further studied for probiotic use and as starter for industrial applications.
