Abstract
The present study mainly covers the purification of selected biomolecules employing unit operations adsorption and aqueous two phase extraction. Experiments were carried out to standardize the process parameters for separation and purification of target biomolecules from natural sources that are often complex mixture. Major emphasis was given to develop a systematic approach for the extraction and purification of anthocyanins from natural sources. Adsorption of anthocyanins was carried out for the highest recovery and stability compared to the conventional purification methods. Dynamic adsorption and desorption process parameters were standardized. Adsorption isotherm and kinetic studies were carried out to understand the mechanism of adsorption of anthocyanins. Adsorption was explained best by using Langmuir isotherm. Structural stability of purified anthocyanins was confirmed using HPLC-MS/MS. Similarly, aqueous two phase extraction was carried out for the purification of anthocyanins and process parameters were standardized. Multistage aqueous two phase extraction was carried out further to increase the purity of anthocyanins. HPLC-MS/MS was carried out to confirm structural stability of the purified anthocyanins. Further, these unit operations were integrated with membrane processes osmotic membrane distillation/membrane pertraction and forward osmosis/direct osmosis to increase the overall productivity of the process. These integrated methods were compared with the conventional methods with respect to stability, recovery and purity. Stability of anthocyanins (obtained by different methods) was evaluated with respect to pH and temperature. The present protocols, although studied for particular biomolecules, can be applied in general to any other biomolecules. The thesis contributes to applied science in terms of process development for the extraction, purification and concentration of selected enzymes from natural complex mixtures.
