Abstract
Background: Dictionary defines pesticide as chemical or biological substances used for destroying harmful insects. Even though they play a significant role in agriculture, they are the most important environmental pollutants due to their wide spread presence in water, soil, atmosphere and agricultural products. They pose a major threat not only to living organisms but also to environment specially ground and surface water; they have lot of hazardous effects, thus it is imperative to detect them in the environment. This study focussed on the detection of two organophosphates methyl parathion and fenthion respectively. Four different techniques, chemiluminescence technique based on luminol-hydrogen peroxide system, Dot ELISA (Enzyme Linked Immuno Sorbent Assay), immuno assay using ELISA and spectrophotometric method were used. Results: Chemiluminescence based method could detect methyl parathion in the range of 500ng/mL to 1mg/mL, while fenthion could be detected in the range of 1mg/mL to 10mg/mL. This method was tested on various environmental samples such as water, soil and vegetable samples. It gave a good recovery as compared to gas chromatography. Dot ELISA method was found to detect fenthion, but could not differentiate between closer concentrations, methyl parathion did not give good results with this approach. The chromogenic method could detect upto fg level. Spectroscopic method could detect methyl parathion in the range of 10μg to 1mg/mL. Discussion: Of the four methods tested here, chemiluminescence based method is found to be more suitable from onsite application point of view owing to its simplicity, rapid detection, sensitivity and the requirement of less reagents. ELISA and Dot-ELISA are based on the antigen-antibody interactions, but they require more time than other assay methods adopted. Spectroscopic method has less sensitivity and can be used only for methyl parathion and hence is restricted for wide scale applications.
