Abstract

Supplementation of food with prebiotics may have a profound effect on stimulation of growth of beneficial bacteria such as lactobacilli and bifidobacteria. Arabinoxylan (AX) being one of the main dietary fiber components in the human diet could function as probiotic was the focus of the present study. Prcbio tic potency of alkali extracted and purified arabinoxylan derived from finger millet bran viz., barium hydroxide (BE) extracted and potassium hydroxide extracted (KE1), with varying molecular weights - 480 kDa and 1080 kDa, respectively, was investigated using individual and mixed cultures of lactic acid bacteria (Lactobacillus brevis 01, Lactobacillus acidophilus Oil, Lactobacillus casei 017, Lactobacillusplantarum 020, Lactobacillus fermentum 156) and bifidobacteria (Bifidobacterium biftdum 235, Bifidobacterium adoiescentis 236). Fermentation of AX was indicated by a decrease in pH (upto4.9), an increase in turbidity (uptol .4 ODat 600nm) and the dry cell mass (2 mg) of all culture filtrates. In addition, the cell wall-degrading enzyme activities such as the xylosidase, arabinofuranosidase and ferulic acid esterase activities of the individual and the mixed cultures were detected in the culture filtrates. The mixed cultures of the lactobacilli and the bifidobacteria grown on both BE and KE1 showed relatively higher activities than all the individual bacterial strains tested. Acetate was the most preponderant short-chain fatty acid (SC'FA) moiety in all the culture filtrates whereas the mixed culture filtrates ofboth the lactobacilli and the bifidogenic bacteria liberated very low amounts of propionate and butyrate. Most of the strains of the lactobacilli and the bifidogenic bacteria grown on BE and KE1 showed bacteriostatic activity towards the food borne pathogenic cultures of Bacillus cereus F 4433 and Escherichia coli MTCC 118. The major importance of the present in vitm studies is that, the purified arabinoxylan isolated from finger millet bran can be considered as potential prebiotic substrate.
