Additional Information: | PHA, bacterial storage polyesters accumulated under unbalanced growth conditions,are considered as environmental friendly, biodegradable, thermoplastics and elastomers that can be obtained from renewable resources. PHA production studies were done with Alcaligenes latus MTCC 2309 in carbon source like Mauha flower and Bacillus cereus “CFR04” in starch medium. The amount of PHA produced by Alcaligenes latus was less when compared to that of Bacillus cereus. Further analysis by NMR proved that the PHA produced by B. cereus is a co-polymer, suggesting its further exploitation in large scale PHA production. A.latus phaC gene of 1.6 Kb which is used for PHA production was efficiently cloned into the Bacillus expression vector pDR67 for further use. The clone was confirmed by restriction digestion, using Bgl II and Hind III enzymes resulting in the release of 1.6Kb phaC fragment from the recombinant vector, suggesting molecular construction of pDRphaC molecule. Tools of bioinformatics were used to construct recombinant map, translation of reading frame and MSA studies. |
