Abstract
Response surface methodology based on a 3-variable, 5-level central composite rotatable design matrix was used to optimize the synthesis of the flavour compound isoamyl propionate by esterification of propionic acid using an immobilized lipase (triacylglycerol lipase, EC 3.1.1.3; Lipozyme IM-20 (Novo Nordisk)) from Rhizomucor miehei. The influence of selected variables, namely, enzyme/substrate (E/S) ratio, substrate concn. and temp. was investigated. E/S ratio and temp. had a strong effect on esterification, while substrate concn. did not. Reaction time did not show a significant effect on ester yield, and conversions were completed in <24 h. The response surface model was found to describe experimental results adequately. Product yield at optimum conditions (24 g/mol E/S ratio, 1.5M substrate concn, 20C) was 1.4M, representing 93% conversion; this yield showed good agreement with that predicted by the model.
