Additional Information: | Hexachlorocyclohexane (HCH) is a chlorinated cyclic saturated hydrocarbon also known as benzene hexachloride, or BHC. HCH is largely used in developing countries like India for the control of agricultural pests and disease spreading vectors like mosquitoes. Even though the agricultural use of tech-HCH is phased in most of the developed countries, countries like India are still using γ-HCH for economic purposes. In addition to already contaminated sites, new sites are being contaminated by γ-HCH and other isomers of HCH. HCH isomers are found to be recalcitrant and persist for a long time in the environment. A microbial consortium capable of degrading tech-HCH was developed by long term enrichment technique. Microbial consortium consists of 10 isolates and all ten isolates were screened for the genes involved in the degradation of HCH. linB and linE genes encode for dehalogenase and dioxygenase enzymes respectively. Pseudomonas diminuta, Pseudomonas fluorescens biovar I, Burkholderia pseudomallei, Pseudomonas putida, Vibrio alginolyticus, Pseudomonas aeruginosa showed positive amplification of linB gene. Pseudomonas flourescens biovar II, Pseudomonas diminuta, Burkholderia pseudomallei ,Flavobacterium , Pseudomonas aeruginosa showed positive amplification of linE gene.Both the genes were cloned successfully into E. coli JM109 using PTZ57 R/T plasmid vector and Sequenced. |
