Abstract

This Dissertation / Report is the outcome of investigation carried out by the creator(s) / author(s) at the department/division of Central Food Technological Research Institute (CFTRI), Mysore mentioned below in this page.

Item Type: Thesis (Masters)
Additional Information: Food and water contaminations, especially by pesticides and foodborne pathogens, are posing a major problem today. The increased use of pesticides and their tendency for bio-accumulation and bio-concentrations is causing widespread ecotoxicity and adverse environmental effects. Moreover they cause chronic and acute toxicity in human beings, not only when consumed but also due to prolonged occupational exposure to treated commodities. Foodborne pathogens and their toxins are leading cause of illness and diseases in the world. They are responsible for millions of cases of infectious gastrointestinal diseases each year. New foodborne pathogens and diseases are emerging everyday because of the pathogen evolution and changes in agricultural and food manufacturing practices. The conventional methods of detection for the pesticide and foodborne pathogens are lacking in terms of sensitivity, specificity, reproducibility and speed. The tests currently used for their detection require lots of time, expensive and complicated equipments with skilled experts. Nanoparticle based biosensors are proving to be the solutions to all these problems. The biosensors are proving to be favourable tools for screening of food contaminants like pesticide residue and foodborne pathogens and their toxins because of their high sensitivity, selectivity, simplicity, reliability and applicability. Moreover they can be used for online or on-field applications, results can be obtained in very short time and the systems are fullyautomated and hence are less labourious. Gold nanoparticles are the so far considered to be the most versatile and the most extensively studied nanoparticles followed by semiconductor quantum dots. Gold nanoparticles are being widely used in bio-assays 132 133 because of their unique photo physical properties like surface plasmon resonance and enhanced light scattering. They can be easily synthesized in a wide range of sizes and shapes and their surface can be tailored by ligand functionalization with a variety of chemical and biomolecular ligands. Modulation of their physicochemical properties and their robustness makes them excellent for use as optical labels in bio-assays. Quantum dots are mainly popular as fluorescent labels as they are ideal donors in FRET and have longer fluorescence life-time and resistance to photobleaching. QDs are being used for biosensing in microarray systems. The spectral properties of QDs are being successfully applied for the multianalyte detection of DNA, proteins, pesticides etc. Hence gold nanoparticles and quantum dots based detection systems are bringing about a revolution in the field of food and environmental safety and analysis.
