Additional Information: Cancer is a complex disease and the incidence is raising constantly posing threat to human population. Currently although cancer stands number two in causing mortality; first being cardiovascular disease, cancer is expected to bypass heart disease as the leading killer globally by 2020 (American Cancer Society, 2006; www.yourcancertoday.com). As estimated, the global cancer burden doubled in the last 30 years of the 20th century, and estimated to be tripled by 2030. Multi-efforts made in the direction of prevention and cure although is helpful in delineating one or few steps in cancer pathogenicity & cancer management; treating or preventing cancer, still poses challenge, particularly in blocking one of the key events in cancer, i.e., metastasis. Metastasis is a crucial step in cancer pathogenicity and adds severity to the disease mainly because of the entry of cancer cell into a highly manipulative stage. Further, besides the severity of disease spread, melanomas pose additional problems to patients, since they are resistant to the option of treatments – radio/chemotherapy. In the current study therefore, attempt has been made to examine the alternate strategies to fight out melanomas. Melanomas since generate enormous amount of melanin, a marker of melanocytes, a key enzyme responsible for production of melanin pigment has been addressed. Since tyrosinase (EC No. 1.14.18.1) is the rate limiting enzyme, investigation was focused on looking for tyrosinase inhibitor from dietary sources. Previously examined dietary pectic polysaccharides from several sources which showed anti-metastatic property by virtue of inhibiting galectin-3 mediated metastasis have been screened for tyrosinase inhibition. Study also addressed to purify and characterize this enzyme for potential application of this enzyme to screen for anticancer compounds. The blockade of tyrosinase enzyme may function like a potential target for management of melanomas in experimental animals with the hope that results can be extrapolated to in vivo human disease condition.
