Abstract

The xylo-oligosaccharides (XOS) obtained by enzymatic hydrolysis of water-extractable polysaccharides (WEP) isolated from Bengal gram husk were tested for their mitogenic and co-mitogenic activities as well as macrophage activation ability. The mitogenic and co-mitogenic activities of XOS were followed by MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] assay. XOS did not show any mitogenic and co-mitogenic activities at various concentrations tested towards both murine thymocytes and splenocytes. However, XOS induced nitric oxide (NO) release from murine macrophages in a dose-dependent manner. The magnitude of NO release by macrophages in the presence of XOS was relatively higher at 48 h compared to at 24 and 72 h. The lack of mitogenic and co-mitogenic activities of XOS isolated from Bengal gram husk towards thymocytes as well as splenocytes may be due to either low degree of polymerization or absence of ferulic or uronic acid moieties or acetyl groups.
