Additional Information: We have successfully developed a novel colorimetric aptasensor using gold nanoparticles (GNPs) for sensitive and specific detection of vitamin B12, one of the most commonly deficient vitamin lead to pernicious anemia. Ahighly stable RNA aptamer that bind to vitamin B12 was employed by structural modification at 2’hydroxyl group of ribose to 2’ Fluoro of all pyrimidines indicated in lower case in 35 bases length aptamer (5’ GGA Acc GGu CAu AAc cAc cuc AGu GcG AGc AA 3’) (Lorsch J.R. and Szostak J.W. 1994, Biochemistry, 33:973-982). Aggregation of GNPs was specifically induced by desorption of the vitamin B12 binding RNA aptamer from the surface of GNPs as a result of the aptamer target interaction, leading to the color change from red to purple. The limit ofdetection of vitamin B12 was100 ng/ml by successful optimization on the amount of the aptamers, GNPs, salts, and stability of aptamer. This colorimetric aptasensor is advantageous over the other conventional methods, in terms of simple signal generation and detection with naked eye, which can be realized in onsite detection for other environmental and clinical samples.
