Abstract

Effect of different level of protein in the diet on the toxicity of erythrosine, carbofuran and nickel was studied. The present study with nickel sulphate has shown that the effect of nickel sulphate at 1000 ppm level in the diet affected the growth of weanling rats more at low protein diet than at high protein diet. A decrease in weight of kidney and liver of rats fed with low protein diet and nickel sulphate indicated that the effect of nickel sulphate is more at low protein diet and decrease in weight may be related to the accumulation of nickel in these organs which had affected the normal synthesis of tissue protein. Histopathological examination of the kidney and liver of the experimental animals indicated the level of toxicity of nickel at different percentage of protein in the diet. Changes in the level of serum constituents like total protein, albumin and globulin and cholesterol is due to nephrotoxicity, hepatotoxicity and cytotoxicity of the metal. Significant increase in creatinine with decrease in urea content of serum shows the effect of nickel on protein metabolism in kidney and liver. Increase in activity of enzyme GOT and GPT in the serum with decrease in activity of enzyme in the liver indicates the malfunctioning of the liver due to damage caused by nickel in the diet. Increase in nitrogenous end product of protein metabolism in urine shows the severity of neptrotoxicity of nickel sulphate in the body at different levels of proteins. Decrease in level of protein in liver with increase in cholesterol level of liver reveals the toxic effect of nickel sulphate on protein synthesis, and degeneration of the hepatocytes resulting in accumulation of cholesterol. Level of micronutrients in serum indicate the effect of nickel sulphate is more on minerals at low protein diet than at high protein diet. Decrease in the level of minerals in the urine of rats fed with low protein diet with nickel sulphate confirms that the oxicity of nickel sulphate is more at low protein diet and protein plays an important role in altering the toxicity of xenobiotics. Excretion of nickel is more in faeces compared to urine and the excretion of nickel is more at low protein diet compared to high proteins diet indicating the effect of protein in nickel toxicity. Nickel sulphate in the diet at 1000 ppm had altered the level of vitamin in the urine indicating the effect of nickel on vitamin.
