Additional Information: Isolation of Tech- HCH – dehalogenase from individual isolates of tech – HCH degrading bacterial consortium was studied. All the ten bacterial isolates of the defined microbial consortium has been found to be necessary for the degradation of HCH. (Manonmani et al 2000). The consortium has been shown to efficiently degrade all the four isomers of technical HCH in water under aerobic conditions (Murthy and Manonmani, 2007). The microbial consortium was induced with both individual HCH isomers and also the technical grade HCH at 10 ppm level. The Cell free extracts of induced cells were studied electrophoretically to find out the number of proteins that might be involved in the degradation of tech-HCH. The γ-HCH induced consortium showed two extra bands of 17kDa and 28kDa molecular weight. The β-HCH – induced microbial consortium showed two extra band of 32kDa and 56kDa molecular weight. Similarly the α-HCH and δ-HCH induced microbial consortium also showed respectively two bands of 40kDa, 64kDa and 17kDa, 46kDa molecular weights respectively. However the technical HCH-degrading consortium, after induction with tech-HCH, showed only three bands of 44kDa, 30kDa, and 17kDa molecular weight (fig.1). However the number of bands were not proportional to the number of enzyme proteins required for the degradation of all four isomers of tech-HCH. The molecular weights of these components were also not in correspondence to those obtained from individual isomers of HCH. The protein band corresponding to 17 kDa was observed in tech-HCH, δ-HCH and γ-HCH induced cells. α–HCH and β–HCH induced cell did not show 17kda band.
