Additional Information: Packaged drinking water has been an unavoidable beverage nowadays among people. In many countries, packaged drinking water is regulated under ‘Food’ and there are many regulations for packaged drinking water worldwide for both microbiological and chemical parameters. In the regulations laid down by USFDA, bacteria should be absent in 100 ml of drinking water. In the regulations of European Union and Codex Alimentarius,bacteria should be absent in 250 ml of drinking water. In India, the regulations are laid down by PFA Act and Rules, which also suggests that in 250ml of packaged drinking water, bacteria should be absent. An attempt was made to study the survival rate of pathogenic bacteria in packaged drinking water. Four bacterial strains were used for the study – Escherichia coli, Pseudomonas aeruginosa, Salmonella typhimureum and Staphylococcus aureus. The bacterial cultures were prepared and compared with the McFarland standard no.1 and inoculated into water samples. Then the water was kept for incubation at two incubation temperatures ie, 35ºC and 22ºC. Aliquots of the sample was taken at periodic intervals and serially diluted and plated onto Plate Count Agar and incubated at the two incubation temperatures. From the colony count obtained, the log counts were determined and graphs were plotted which depicted the survival profile of the four organisms in packaged drinking water. The chemical parameters of the water sample was also analysed which included Total dissolved solids, Chlorides and Alkalinity.
