Additional Information: | <p align="justify">The Biopolymer is assuming importance because of the pollution problems associated with synthetic plastics, which are produced from non-renewable energy sources and are non-biodegradable. As an alternative to this, Polyhydroxyalkanoates are gaining much attention in recent years as they posses similar properties to conventional plastics and are completely biodegradable. Polyhydroxyalkanoates are microbial thermoplastics, which are regarded as potentially useful polyester replacing petroleum derived thermoplastics. Polyhydroxybutyrate (PHB) is the best-known member of the PHA series of polyesters. The sequence homogeneity within the species of 16SrRNA makes the sequencing and analysis of 16SrDNA as the best method for identification of microbial strains at the species level since phenotypic characters may not precisely differentiate species. With this background the following objectives i.e. PHA production of organism obtained from the project culture collection of Food Microbiology Department, CFTRI, Identification and molecular characterization of PHA producing organism and Detection of PHA Synthase gene was studied. The organism was observed to be gram-positive rods. The Bacillus sp was found to produce an average of 62.18% Polyhydroxyalkanoates. The molecular characterization using 16SrRNA and phylogenetic analysis proved that the organism belongs to genus Bacillus and species flexus. The detection by PCR method confirmed that the organism Bacillus flexus contains phaC gene and thus concluded that the organism is a potent producer of Polyhydroxyalkanoates.</p> |
