Additional Information: | Food industry has been using polysaccharides of different origins to improve the rheological characteristics of various food products. Earlier, most of the biopolymers used were of plant origin. These are being replaced by microbial biopolymers which has several advantages over the biopolymers obtained from plant origin. Microbial biopolymers have found a place in food industry as texturizing, binding, flavor enhancing, viscosifying, stabilizing agent. These biopolymers are making a foray into other industries including pharmaceutical and cosmetic industries. Its potential applications have necessitated the need for a better understanding of their isolation, production and optimization. The project was focused on the optimization of various parameters (both nutritional and physical). The log phase of Leuconostoc sp. CFR 2181 was determined to be between 2.5 hrs to 6.5 hrs, which helped in deciding the duration of the incubation while preparing the inoculum.Effect of rate of inoculum, there was a change in inoculum rate from 1 to 10% (v/v) did not make any marked difference on the biomass content. Although increased production of EPS with the increase in the rate of inoculum from 1 to 5 % was noticed, further increase to 10 % (v/v) decreased the EPS content. It was concluded that the rate of inoculum at 5 % (v/v) level could be used for optimum production of EPS. Other effects studied include: addition of media every 2 hrs; different sources of carbon; different concentrations of sucrose as carbon source; period of incubation; different initial pH; neutralization of pH; temperature in relation to static condition; kinetics on EPS production. |
