Effect of feeding sesame oil and groundnut oil on iron induced hepatotoxicity and lipid peroxidation in rats.

Abstract

The protective role of sesame oil against Fe induced lipid peroxidation and hepatotoxicity was investigated using an animal model. Male Wistar rats were fed sesame oil (SO) or groundnut oil (GNO) for 8 wk. Half of the rats were injected with 30 mg Fe2+(as FeSO4)/kg body wt. and half were injected with saline (control). Basal and incubated lipid peroxidation levels in liver homogenates and microsomal suspensions were measured. In rats injected with Fe and fed SO, lipid peroxidation was lower in serum, liver homogenates and liver microsomal suspensions compared to those fed GNO. Superoxide dismutase (SOD) activity was less in SO fed rats as compared to GNO fed rats. No difference was observed in SOD and catalase of GNO and SO fed rats though a significant difference (P < 0.001) was found in rats injected with Fe2+. Alanine amino transferase (ALAT), aspartate amino transferase (ASAT) and lactate dehydrogenase (LDH) levels were lower in the serum of the SO fed group. Results indicated that feeding SO lowered lipid peroxidation by enhancing antioxidative enzyme activity and also decreasing activities of ALAT, ASAT and LDH, suggesting that hepatic injury can be reduced by inclusion of SO in the diet.
