Additional Information: Chicken intestine is an important source of proteases, particularly alkaline proteases. Aqueous-two phase consisting of a polymer and a salt has been used for separation and purification of various enzymes. This study was carried out with an objective of evaluating the effect of different parameters of aqueous two phase system such as different salts, salt concentration, protein concentration, temperature, PEG concentration and pH on the partition behaviour of protein and alkaline protease from chicken intestine. The results of the study indicated that the partition behaviour of protein and alkaline protease from chicken intestine is influenced by the type of salt used in aqueous-two phase system with a two phase system consisting of PEG-6000 and sodium citrate or potassium phosphate showing higher enzyme recovery and enzyme purification. Salt concentration in the system also affected the protein and enzyme partition with sodium citrate at 15% level showing better enzyme recovery and purification. The partition of protein and enzyme was also influenced by the level of protein in the system with lower level of protein in the system showing desirable results. The enzyme yield and purification was also affected by the temperature at which the ATPS is performed with lower temperature showing better results. The study indicated that ATPS system consisting of PEG 6000 and sodium citrate can be effectively used for separation and purification of alkaline proteases from chicken intestine. However, as multiple factors influence the behaviour of protein and enzymes in an aqueous-two phase system, it is necessary to evaluate the combined effect of all the factors involved. Once the major factors influencing the system are identified, it is desirable to optimize the conditions necessary for the effective separation and purification of alkaline protease from chicken intestine using ATPS and to conduct scale-up studies.Chicken intestine is an important source of proteases, particularly alkaline proteases.
