Additional Information: | A study was conducted on the application of ammonium sulphate precipitation technique for purification of acidic proteases from chicken intestine. Acid protease can be obtained from chicken intestine by applying simple and effective method of ammonium sulphate precipitation. The sample was subjected to different saturation from 40%-80% and the protein fractions were collected at each level. Highest purification was obtained in 40% saturation, which partially purified enzyme that can be used in tannery industry, fermentation industry, meat tenderization, baking industry and in diary industry. Characterization of partially purified acidic protease showed optimum pH of 4, temperature optimum of 400C, which showed stability even after incubating for 90 minutes. The activity can be enhanced using metal ions such as ZnSO4, CaCl2, and MgCl2. Molecular weight of the partially purified enzyme ranged from 27KD- 97KD. Proteases make up to 48% of industrial enzymes and are mostly used in detergents, leather production and food industry. Therefore extraction and purification of acidic protease is necessary which not only retains the valuable biomolecules but also avoid pollution of environment that is caused due to the byproducts obtained from poultry industries. |
