Additional Information: Lipid oxidation is common metabolic reaction in all biological systems, both appearing in developmentally and environmentally regulated processes and as response to biotic and abiotic stresses. Lipoxygenase (LOX) are a family of non-heme iron containing dioxygenases. They catalyze the insertion of molecular oxygen into poly-unsaturated fatty acids that contain one or more pentadiene moieties to yield the corresponding hydro-peroxides. LOXs are present in animals and plants where they catalyze the oxidation of polyunsaturated fatty acids (PUFAs) and play a key role in the production of signaling compounds and in the degradation of cellular and intracellular membranes during development. A number of diseases in humans, different types of cancer and neurodegenerative diseases, have been related to an increased level of LOX activity. Besides the pathological role of these enzymes, LOX is initiator of uncontrolled oxidation processes that are one of the main causes of food quality decay. LOXs are responsible for the development of rancidity in fat-containing foods. Generation of off flavors, loss of nutritional value and loss of economical value of food products are the result. Due to these adverse effects associated with the activity of these enzymes, inhibition of LOXs is an important target in both agricultural and pharmaceutical industries. A trial has been made to screen and produce an inhibitor of lipoxygenase from fungal sources. The present work deals with the isolation, characterization and purification of lipoxygenase inhibitor from fungal origin.
