Additional Information: In this literature, the work is done on the study of microbial diversity using the metagenomics approach, which refers to the method of directly recovering the genetic material from the environmental samples. Here the sample was taken from soil of different areas such as soil from the rhizosphere region, from the site of garbage dumps and sewage and burnt dump areas. From these mentioned soil samples the analysis of the probable presence of microorganism particularly the bacteria are carried out. The DNA were isolated from the soil samples and these were quantified by using agarose gel electrophoresis, this enabled the qualitative measurement of the DNA of the given sample loaded in the wells and their intensity which is also a measure indicting the yield and its purity of the genomic DNA. This obtained DNA were further amplified by the technique of PCR ( Polymerase chain reaction), for this purpose universal primers were used instead of specific primers since the diversity or similarity of the microorganisms were not known. The combinational use of different primers enabled the amplification of the sample, though for not all of them, amplification was not possible owing to the nature and sequence of the primers used with the corresponding 16 sDNA and it’s complementary DNA sequences. Again the PCR amplicons were run through agarose gel electrophoresis unit with a standard DNA ladder to compare the size of the DNA fragments based on their molecular weight and density. Finally from the results obtained they were compared for the similarity between the bands of DNA, which directly refers to the total population of the microorganism of that particular soil sample. Based on this, further study can be made depending on the findings a suitable set of techniques can be carried out to know in depth the phylogeny between the species of microorganisms present, which in this case refers to the prokaryotic microorganism like the bacteria.
