Additional Information: Age related Macular Degeneration is a physiological condition affects the acuity of the central vision due to the degeneration of macula the central region of the retina owing lower levels of antioxidant carotenoids lutein and zeaxanthin. Under such condition, enhancing lutein bioavailability is vital for which newer techniques like encapsulation rather dietary means may advantages. Hence, the objective of the work was to prepare and characterize nano-encapsulated lutein followed by bioavailability of lutein in vitro. Lutein was extracted and purified from the marigold flower and used for encapsulation. Nanoencapsulation of lutein was done based on chemical methods. After screening of different polymers such as sodium alginate, chitosan, PLGA, HPMC, gelatin and gum acacia to encapsulate lutein, PLGA was selected for further studies. The PLGA nanoencapsules were prepared based on the sonication and solvent evaporation technique. The encapsulation efficiency of the PLGA nanoencapsules was found to be 85%. The SEM analysis of the nanoencapsules showed the size of particles < 300nm and were spherical in shape with moderate uniformity. The particle size analysis showed the size of nanoencapsules ranging from 100 to 600nm. AFM analysis revealed that the size of the nanoencapsules were more or less 350nm and the three dimensional view of the surface of the nanoencapsules were also obtained. The FT-IR analysis of nanoencapsulated lutein further confirmed the presence of both PLGA and lutein in the nanoencapsules. It was found that the bioavailability of lutein from nanoencapsulated was higher than from mixed micelles and standard lutein dispersed in groundnut oil by the in vitro digestion method. The everted sac analysis of lutein showed the absorption through the intestine was higher in the case of nanoencapsulated lutein compared to that of mixed micelles. The anti-oxidant assay showed that the percentage of radical scavenging activity of the nanoencapsulated lutein was slightly lowers then the standard lutein. From the results it is concluded that nanoparticles loaded with a fat-soluble lutein can be prepared with a synthetic polymer PLGA. Further, the biological availability and antioxidant property of lutein from nanoparticles was significantly higher demonstrating future application of this technique in the area for ophthalmology to meet ARMD.
