Abstract

Chitosan is a linear polysaccharide composed of β-(1-4) linked N-acetyl glucosamine and D-glucosamine. Chitosanase is an enzyme that hydrolyzes chitosan and exclusively produces chitosan oligosaccharides (COS) as the end product. The COS generated from chitosan by enzymatic methods possess numerous biological activities. In this study, the molecular identification and production of chitosanase from a soil fungus (CFRNT16) was described. The organism (CFRNT16) was screened as a chitosanase producing fungus by observing the zone of clearance around the colony on a colloidal chitosan agar medium. On the basis of the partial sequencing of 18S rRNA gene, the organism CFRNT16 was identified as Mucor racemosus. After the identification of fungus, the chitosanase production by submerged fermentation (SmF) was performed. The maximum enzyme activity obtained after 36 hours of cultivation was 22 (U/ml) in a colloidal chitosan medium and 20 (U/ml) in a shrimp shell powder medium. The antioxidant property of fermentation broth was also established. The result of the investigation confirmed the potential of M. racemosus (CFRNT16) for the production of commercially important chitosanase.
