Abstract
A simple protocol was developed for purification of the alpha-, beta- and gamma-kafirins (alcohol-extractable proteins) of sorghum based on extractability differences. The relationship between kafirins extractable in 60% t-butanol and cross-linked kafirins extractable in 60% t-butanol/0.6% 2-mercaptoethanol was examined using immunochemical techniques. The immunological relationship between kafirins and prolamins from other tropical millets and cereals was also investigated. Polyclonal antibodies were raised against the kafirins; immunoblotting showed no cross reactivity between the 3 kafirins. N analysis, SDS-PAGE, immunoblotting and ELISA demonstrated the quantitative differences between the kafirins and the cross linked kafirins. ELISA determined that the increased extractability of beta- and gamma-kafirins was responsible for the increase in total kafirin extracted on addition of a reducing agent to aqueous t-butanol. Homologies were thought to exist amongst the prolamins contained in tropical millets and cereals as all 3 kafirin antibodies reacted with the specific prolamins.
