Additional Information: | TThe present study is an attempt to identify the organisms involved in the pediocin production isolated from different sources by RAPD PCR. LAB were isolated from different sources using selective media MRS. The bacteriocinogenic LAB were screened by presence of inhibition zones against the indicator organism, Listeria murrayi FB69. A total of 23 culture isolates were obtained from the source samples: beans, chicken intestine, fish intestine, idli batter, and soya. The putative culture isolates with pediocin production was identified by pedB amplification through PCR. The soya and idli batter isolates gave positive pedB amplification. The culture isolates were further characterized by using a PCR based methodology called RAPD, where the native isolates profile was compared to the positive pediocin producing Pediococcus acidilactici K7 and non pediocin producing Enterococcus faecium C20. The isolates S1, S5, IB2, IB3, C1, B3 gave a similar banding pattern to that of C20 and hence are all pediocin producing Enterococcus faecium. The isolates S2, S3, S4 gave similar banding pattern to K7, indicating they are Pediococcus acidilactici. The other isolates, S6; IB1; IB4, IB5, IB6; IB7; C2 gave a similar banding pattern and different from K7 and C20; hence are pediocin producing organisms other than Pediococcus and Enterococcus, as they gave pedB PCR amplification. Based on RAPD results, nine culture isolates (S1, S3, IB3, IB4, IB7, B3, B1, C1, FI) were selected for further characterization. On testing the inhibitory substance present in the culture filtrate, the antimicrobial compound from the isolates was observed to be proteinaceous in nature, heat tolerant, and pH stable. Microbial tests and biochemical tests indicated that cells are Gram- positive, cocci shaped, homofermentative, and catalase negative. The results suggests that the native isolates S2, S3, S4 from soya are Pediococcus, and other native isolates are pediocin producers which are different from the control, Pediococcus acidilactici K7. |
