Additional Information: | Yersinia, E.coli and Streptococcus are the most common causes of food borne diseases worldwide. Food poising is caused by the ingestion of food contaminated with toxin produced by Yersinia, E.coli and Streptococcus. Traditional method of detection by plate count method followed by biochemical tests is time consuming and labour intensive. Hence, for the rapid detection of toxigenic species the molecular methods have been developed. PCR requires thermocycler followed by electrophoresis detect the pathogenic bacteria by amplified products. So a novel nucleic acid amplification method based on loop mediated isothermal amplification(LAMP) was studied to detect the Yersinia, E. coli and Streptococcus in food and water samples. The aim of this study was to develop a method to detect pathogenic bacterial species in contaminated food and water samples at field level In our studies, Yersinia, E.coli and Streptococcus were isolated from contaminated food samples on selective agar plates. These were screened by biochemical tests and 16SrRNA to confirm their identity. These were subjected for molecular detection such as PCR and LAMP method. The results of the LAMP assay completely compared with PCR assay. The isolates which gave no amplification in PCR were found to give amplification by LAMP. Matrix effect was not observed in LAMP method with different food matrices. LAMP method rapidly detected toxigenic strains within 60 minutes. This method is found to be suitable for food safety applications. |
