Abstract

Here flaxseed, a storage protein has been fractionated into albumin, globulin, glutelin and prolamine. Fractionation was carried out from defatted dehulled flaxseed meal by sequential solvent extraction. Globulin and albumin fractions were the predominant protein fractions accounting for 47.3 % and 37.8 % respectively, while the glutelin and prolamin accounted for 16.7 % and 9 % respectively. The current study is focused on isolation of major fraction of flaxseed protein (globulin isolate) with high protein content and its evaluation for cholesterol lowering effect in hypercholesterolemic induced rats. The maximum extractability (51.45 %) of flaxseed globulin was showed at 3 % sodium chloride (NaCl) concentration. Flaxseed globulin showed maximum solubility at pH 2 and the isolelectric point of globulin was at pH 5. The globulin isolate prepared had a protein content of 93 %. SDS PAGE patterns of the globulin isolate showed three bands at 36.5 kDa, 26.6 kDa and <26.6 kDa. In vitro protein digestibility of globulin (92.6 %) indicated that the protein is easily digestible. The animals were divided into four groups and fed with four different diets; casein control (CT), casein high cholesterol diet (HCD), flax control (CT) and flax high cholesterol diet (HCD) for five weeks. Hypercholesterolemia was induced using 1 % cholesterol. The lipid profile of both plasma and liver sample showed no hypocholesterolemic effect on rats fed with flax diets. Our results reveal that globulin is the major fraction of defatted flaxseed meal and this globulin has no effect on lipid profile of hypercholesterolemic rats. The globulin isolate maintained the levels of ascorbic acid and lipid peroxidation in hypercholesterolemic induced rats.
