Additional Information: A study was conducted to isolate high quality RNA by grinding whole banana pulp, vanilla leaf and algae by three different methods viz. Phenol:Chloroform, NaCl method and Ambion kit. All the protocols were found to be suitable for the isolation of total RNA from all the samples. However, if large quantities of RNA are desired, it is advisable to use NaCl since this method produces significantly larger amounts of RNA than the Ambion kit protocol and it was found to be easily scalable. Several novel genes was identified related to fruit ripening whose action in the ripening process was not obvious earlier. An mRNA differential display approach was followed to identify unreported genes from banana. Most of these genes are fruit specific and show differential patterns of expression during the course of ripening. An attempt was made to dissect the ethylene response from the ripening response.
