Abstract
3-methylglutaryl coenzyme A reductase (HMGCR) is a regulatory cholesterol synthesis enzyme in the mevalonate pathway. Statins are the major synthetic drugs as inhibitor of HMGCR but it’s associated with adverse effect so it is advisable to provide alternative therapeutic candidate for cholesterol homeostasis. The current investigation indicates the cholesterol-lowering effect of Pigeon pea by-product protein hydrolysate (PBPH) fraction by inhibition of HMGCR and cholesterol micelles. The PBPH fractions control the cholesterol levels by two ways, first, PBPH fractions (> 30 kDa and > 10 < 30 kDa) significantly block the recycling of bile salts to the hepatocyte cells with direct excretion via the digestive system. Second, PBPH fraction (< 10 kDa) exhibits inhibition of cholesterol micellar, as well as the catalytic domain of HMGCR in a dose dependant manner. The inhibition of HMGCR has been verified through HMGCR protein expression in the cholesterol synthesis pathway. The HMGCR protein shows significant up-regulation upon treatment with PBPH fraction < 10 kDa in HepG2 cells. The PBPH fraction shows potent inhibitory capacity on cholesterol homeostasis suggesting it to have hypocholesterolemic properties. It could find an admirable application for the development of functional foods.
