Abstract

This Dissertation / Report is the outcome of investigation carried out by the creator(s) / author(s) at the department/division of Central Food Technological Research Institute (CFTRI), Mysore mentioned below in this page.

Item Type: Student Project Report
Additional Information: The peptide OB-23 was synthesized using CS136 peptide synthesizer using Fmoc strategy. After synthesis the peptide was cleaved from the resin using a cleavage mixture consisted of 8.75ml TFA, 0.5mlwater, 0.5mlEDT, 0.25ml thioanisole. The peptide was precipitated using diethyl ether. The peptide was analyzed by ESI-MS and analytical RP-HPLC which showed the presence of exactly half the mass of the peptide i.e.1259.14Daltons corresponding to m/2,the mass of the peptide is 2516.85Daltons.The so analyzed peptide was purified by preparative RP-HPLC and the fractions collected for peaks were subjected to ESI-MS analysis. The fractions that showed similar mass were pooled and lyophilized. The pooled fractions from each peak were crosschecked for purity by analytical RP-HPLC. In summary a 23 residue peptide OB-23 was synthesized and purified to homogeneity.
