Abstract

Chickpea husk rich in non-starch polysaccharides (NSP) were extracted sequentially using cold water, hot water, 0.5% ammonium oxalate (AO) and 0.5% EDT A in 0.9, 0.4, 4.4 and 3.8% yields, respectively . Sugar composition of both AO and EDT A extract revealed the preponderance of galactose, xylose, arabinose and smaller amounts of rhamnose and mannose. Ammonium oxalate and EDT A extracts consisted of 71.2 and 62.4% uronic acid respectively , which is comparatively higher than that present in other dietary fibre components isolated from chickpea husk in the present study . Ammonium oxalate and EDT A extracted fractions which are rich in pectins were acid hydrolyzed to obtain pectic oligosaccharides. These oligosaccharides were detected by TLC and analysed by LC-MS. The antioxidant activity of pectic oligosaccharides derived from ammonium oxalate showed significant DPPH radical scavenging activity compared to the EDT A derived pectic oligosaccharides and can be explored as novel antioxidant. The prebiotic properties of both AO and EDT A derived pectic oligosaccharides were studied using Lactobacillus acidophilus (NDRI 011) and Bifidobacterium adolescentis (NDRI 236). Pectic oligosaccharides derived from the pectin isolated by EDT A extraction had relatively higher prebiotic activity than that derived from AO extracted pectin
