Abstract
The mechanism of citric acid production by a selected isolate of Aspergillus niger-16 was followed by monitoring different enzymes of Krebs cycle when the fungus was cultivated in defined media containing alcohols, fluroacetate and coconut oil. There was two-fold increase in citrate synthase activity in the presence of alcohol, while aconitase activity was reduced to a quarter. Fluroacetate completely inhibited aconitase activity and significantly altered activities of other enzymes also. Coconut oil did not influence the activities of any of the enzymes of the citric acid cycle. All the concerned enzymes were present during accumulation of citric acid.
