Additional Information: Fusarium belongs to a large group of mycotoxigenic filamentous fungi that are pathogenic to plants, animals as well as human beings. They produce a potent toxin Trichothecene, which has harmful effects on the food grains when infected with Fusarium spp. Detecting the production of trichothecenes is a laborious process which involves Thin Layer Chromatography (TLC) and High Performance Liquid Chromatography (HPLC).The main aim of this project work was to device a method to easily detect trichothecene production. Reporter gene like Green Fluorescent Protein (GFP) gene was used to tag the tri5 promoter of F.cerealis, which codes for trichothecenes. Cloning of this tri5 promoter into GFP vectors pCAMBIA1302 and pCAMBIA1304 (by replacing the CaMV promoter) was tried. Due to technical difficulties, this was not achieved. Agrobacterium mediated transformation of F.cerealis with pCAMBIA1302 and pCAMBIA1304 proved to be simple and successful when compared to electroporation. The transformation was confirmed by the fluorescing of the hyphae when viewed using a fluorescent microscope. This indicated that the GFP gene was taken up by F.cerealis and also expressed, which consequently proved that trichothecene was also produced. By varying the environmental factors in which the fungus grows, the exact period of trichothecene production can be detected using GFP. The GFP may be expressed from the promoter of genes involved in trichothecene synthesis to determine the function of various cis acting elements in the promoter.
