Additional Information: | The inhibition or suppression of rice-bran lipase is a major concern of the rice industry and attempts have been made to chemically inactivate lipase to obtain better quality oil and rice bran fit for food and feed purposes. One of the most successful methods developed in recent years to inhibit gene expression in plants has been the concomitant expression of an introduced antisense gene. Therefore the starting point for an efficient method for stabilizing rice bran would be an attempt to produce an antisense lipase in rice plants. In an attempt to correlate at the molecular level, and restrict the activity of rice bran lipase for the production of a better quality oil and bran the rice bran lipase gene was isolated and characterised with specific temporal and spatial patterns of expression. In this investigation the major objective was to clone the cDNA fragment already available in the laboratory into pRSETC vector, screen the clones for the RBL gene and expression of the protein in E. coli. |
