Additional Information: Lipases (Triacyl glycerol hydrolases ) catalyze the hydrolysis of ester linkages in lipids with release of fatty acids and alcohol. They have vast potential for industrial application in number of industries. For large scale production of lipases , along with other parameters rheology is an important aspect. Rheology is a study of change in form and flow of matter, embracing elasticity, viscosity and plasticity. The scope of this project is to study the rheological project is to study the rheological properties of lipase solution . The objective is to charecterize rheological model of lipase solution and A.niger broth. Study of shear forces on lipase activity and activity recovery in lipases after shear. A number of shake flask experiments conducted at different intervals confirmed Bingham plastic behavior for both lipase solution and A.niger broth. Shear inactivation of lipases was investigated which shows that lipases suffer minimum activity loss at low shear rates and activity loss is high with increasing rate of shear and exposure time. Activity recovery studies done on the lipase suggests activity recovery is possible after removing shear at low shear rates but lipase inactivates at high shear rate and longer exposuretime. Based on this investigation, strategies can be made for improvements in the yield of lipases in commercial production.
