Additional Information: | Lipases catalyze the hydrolysis of ester linkages in lipids with the release of fatty acids and alcohol. The objective of this project was to study the influence of inoculum on lipase production from Aspergillus niger and also to develop a suitable media for the growth of organism. Lipase production in shake flasks and also at the fermentor level was carried out. In continuous series of shake flask experiments with Corn steep liquor media, the maximum activity was found to be 15±2 U/ml. An induction media with tributyrin concentrations of 0.05%, 0.10%, 0.20% were used for lipase production. It was found that 0.10% tributyrin slightly increased the lipase production compared to the control experiments, where tributyrin was not used. The maximum activity observed when spore suspension from 0.05%induction media was 17.62 U/ml. Induction media with 0.20% was found to be inhibitory for lipase production. Shake flask experiment, where inoculum prepared from potato dextrose agar media gave maximum activity of 17.37 U/ml at 48th hour. One fermentor trail was conducted for scale up of lipase production. The highest activity was 7.5 U/ml at 36th hour. Down stream processing by using flash evaporator was carried out to concentrate the enzyme obtained from shake flask experiments. |
