Additional Information: | <p align="justify">Pesticides are one of the major pollutants found in environmental and food samples. Application of organochlorine and organophosphorous pesticides in agriculture has been practiced in many countries. As a result of the increased use of pesticides in agriculture in the last few decades, ground water, raw food materials and processed food are becoming contaminated with pesticide residues. Major health problems occur due to the use of contaminated water and food. Because of the wide and growing use of pesticides, pollution monitoring and protection agencies need rapid and sensitive methods for pesticide analysis. The techniques currently applied for pesticide determination are primarily chromatographic involving various detection principles. The chromatographic methods have sufficient sensitivity for pesticide detection, but they are time consuming and involve expensive instrumentation. Biosensor is an alternative tool to detect the pesticide rapidly and economically. In recent years, immunoassay methods have been increasingly applied to simpler and quicker pesticide detection. Immunochemical techniques are considered as a good alternative to the more classic chromatographic and enzymatic methods. Most of these methods are ELISAs with photometric detection. Till date sensitive (ppt level) and rapid techniques are not available to detect organophosphorous pesticides. An immunosensor based on chemiluminiscence for pesticide detection is being developed at CFTRI. In the present study IgY antibodies were used for specific detection of Methyl parathion and Ethyl parathion. The antibodies were immobilized on sepharose matrix. Flow ELISA was performed using the immobilized matrix, chemiluminiscence assay was done and images were captured using luminometer and CCD camera. A good gradient of light intensity was obtained with different pesticide concentration. The IC 50 value for EP was also determined. The detection system developed here was found to be a highly reliable, fast and sensitive test for detection of organophosphorous pesticides.</p> |
