Abstract

Str. fradiae strain SCF-5, known to produce cell-bound glucose isomerase, also produces small amounts of extracellular xylanase. Spores and fragmented vegetative cells of the strain were subjected to UV radiation in an attempt to produce mutants having increased xylanase-producing ability; vegetative cells were comparatively easy to handle, while spores formed clumps and were difficult to suspend uniformly. A UV dose resulting in death of 95-99% of the strain was usually employed. Capacity of 3 mutants (named UV-1, UV-2 and Grey) with an increased (up to 4-fold) xylanase-producing ability to hydrolyse larchwood xylan was examined. Attempts were also made to cultivate the 3 mutants on xylan-containing substrates (paddy straw, bagasse, wheat bran, sawdust). While, in most cases, a considerable amount of xylanase was produced, little or no cell-bound glucose isomerase was detected. It is therefore concluded that the mutants generated insufficient amounts of xylose to induce glucose isomerase.
