Abstract

2 fractions, designated A and B, having haemagglutinin (lectin) activity were purified from aqueous extracts of field bean seeds by isoelectric precipitation, and ammonium sulphate fractionation. The haemagglutinin activity of fraction A seems to be due to presence of fraction B. Fraction B was homogeneous by acrylamide gel electrophoresis and ultracentrifugation. The mol. wt. of this fraction was approx. 100 000, as determined by gel filtration. Field bean lectin does not need metal ions for its haemagglutinating activity and among a number of sugars tested, D-glucose, D-mannose and methyl-alpha-D-mannopyranoside were inhibitory. Reversible agglutination could be demonstrated with the inhibitory sugars. Erythrocytes from rabbit, guinea pig, rat and fowl were easily agglutinated while human (A, B, O group) erythrocytes were somewhat resistant.
