Abstract

The alkali degradation test developed involves placing 5-6 kernels in a 5-cm diam. Corning Petri dish stranding on a black sheet, covering the kernels with 10 ml of 2% NaOH, covering the dish and leaving undisturbed, preferably overnight. The endosperms are rapidly attacked and completely gelatinized within 3-4 h, the detached residual bran layers standing out very well against a nearly transparent background. The approximate degree of milling is determined from the nature and amount of the separated bran layers by means of a score card. The alcoholic alkali bran-staining test developed involves placing 1 g kernels in a 5-cm diam. Corning Petri dish placed on a white background, and covering the kernels with 10 ml of 2% aqueous KOH:ethyl alcohol (or denatured spirit) (1:2, v/v) solvent. After 15-30 min, the kernel are carefully observed with a magnifying glass and the relative size and amount of the yellow-brown patches or streaks are noted and the degree of milling assessed by means of a score card. Both methods should prove suitable for determining the approximate degree of milling of rice.
