Isolation, enumeration and PCR characterization of aflatoxigenic fungi from food and feed samples in India.

Abstract

The prevalence of aflatoxin-producing fungi and aflatoxins was examined in 54 market samples consisting of 9 different food and feed commodities from Mysore city, India. Results showed that 32 samples were contaminated with aflatoxigenic fungi. The total mycoflora and aflatoxigenic fungi in different food and feed commodities were in the range 0.2-260 and 0-100 cfu x 103/g, respectively. A total of 136 fungi were isolated, 32 of which were Aspergillus flavus strains; 26 of these were aflatoxigenic. Aspergillus differential medium and PCR were used to characterize the A. flavus group of fungi comprising A. flavus, A. parasiticus, A. oryzae and A. sojae. 2 different sets of PCR primers specifically targeted to the aflR and omt genes of the aflatoxin biosynthesis pathway were used. Most A. flavus group fungi reacted positively with the primers to produce amplicons of an expected size; 796 bp for aflR and 404 bp for omt. Groundnut (peanut) and groundnut cake were among the 9 commodities screened for aflatoxins only which were contaminated with aflatoxins B1 and B2. Aflatoxin contamination levels exceeded the Indian regulatory limit of 30 g/kg in these commodities.
