Abstract

Fructooligosaccharides have been produced from sucrose by the action of a microbial fructosyl transferase (FTase). Influence of nutritional parameters on FTase production and of the reaction conditions on fructooligosaccharide production by Aspergillus oryzae CFR 202 were studied using Plackett-Burman design. 16 variables, including 8 medium components and 8 reaction parameters, were studied initially. From this study, KH2PO4 and sucrose contents of the fermentation medium, fermentation time, reaction time and pH of the reaction mixture were identified as important factors and chosen for further optimization. The highest conversion yield obtained was 58% (w/w) based on initial sucrose concn., which was 33% higher than that achieved in the initial screening experiments.
