Abstract

Differences in the composition and distribution of kafirins (sorghum prolamins) extracted from endosperms of hard and soft sorghum (Sorghum bicolor (L.) Moench) genotypes, and differences in the distribution of kafirins in the vitreous and floury fractions within kernels of varying hardness were studied. Kafirins from 7 sorghum cv. were analysed by SDS-PAGE and ELISA. In hard endosperm kernels, ELISA showed that levels of all 3 kafirins (alpha, beta and gamma) were high. In hard kernel vitreous endosperm portions, gamma-kafirin was particularly high. In soft kernels, the ratio of gamma-kafirin to alpha-kafirin was higher in the floury portions. Tissue print immunoblotting showed that in soft kernels alpha-kafirin was fairly uniformly distributed throughout the endosperm, whereas beta- and gamma-kafirins were concentrated in the central floury endosperm portion. In hard kernels, all 3 kafirins were uniformly distributed throughout the endosperm. It is concluded that content and distribution of kafirins within sorghum kernels varies in grains with different endosperm hardness.
