Abstract

Optimization of the downstream processing of C-phycocyanin from cultures of the microalga Spirulina platensis using an aqueous 2-phase extraction process is reported. Effects on the extraction of pH, phase composition and nature and vol. ratio of the phases were assessed. Results show that use of an extraction system comprising polyethylene glycol (4000) and potassium phosphate at a phase vol. ratio of 0.8 and pH of 6 were suitable for extraction of C-phycocyanin from S. platensis. Under these conditions, max. purity was achieved when 3 extractions were performed. Purification of the pigment from the organic extraction solvent could be achieved using ultrafiltration and freeze drying of the purified C-phycocyanin was used to obtain the pigment in powder form.
