Abstract

Weanling male Wistar rats weighing about 40 g were given a diet containing only 8% casein and lacking pyridoxine for 38 days after which they were divided at random into 4 groups. For the next 7 to 10 days 2 groups continued on the low-protein diet, one with a supplement of pyridoxine hydrochloride 1 mg%, and 2 groups were given 18% casein diet, one supplemented with pyridoxine hydrochloride. Two groups were long-term controls, one at each dietary protein level supplemented with pyridoxine. In some experiments the pyridoxine hydrochloride was given by subcutaneous injection on 2 days and the rats were killed 24 h after the second injection. Pyridoxine supplementation significantly increased alanine aminotransferase activity in liver and heart tissue homogenates; the increase in dietary casein to 18% had no effect in the short experiments, but increased the enzyme activity in the long term. Tyrosine aminotransferase in liver supernatant was increased by pyridoxine supplementation, but mitochondrial tyrosine aminotransferase showed no dependence on the vitamin. In rats given a diet containing 2% skimmed milk powder, liver mitochondrial leucine aminotransferase activity was low; when the rats were refed on 10% skimmed milk powder diet, the activity was restored.
