Additional Information: <p align="justify">Capsaicin synthase is the key regulatory enzyme in capsaicin biosynthesis. So far, there are no reports on purification and characterization of this enzyme. The present work was investigated with a purpose of partial purification and characterization of capsaicin synthase from placental tissues of Capsicum annuum var Arka Lohit from IIHR, Bangalore. Capsaicin synthase has been purified to near electrophoretic homogeneity, which was found to be heterodimer with a molecular weight of 53 kDa and 35 kDa. Using different chromatographic techniques, two dimers were separated in active form. 53 kDa subunit was found to be 8 methyl nonanoic acid dehydrogenase activity which biosynthesizes 8-methyl-nonenoic acid whereas 35 kDa subunit is capsaicin synthase. Purified capsaicin synthase was characterized for optimum substrate concentration in terms of km and v-max values. It was also found that capsaicin synthase is a cationic protein as confirmed by DEAE cellulose.</p>
