Abstract

This Dissertation / Report is the outcome of investigation carried out by the creator(s) / author(s) at the department/division of Central Food Technological Research Institute (CFTRI), Mysore mentioned below in this page.

Item Type: Student Project Report
Additional Information: Advances in industrial biotechnology offer potential opportunities for economic utilization of agro industrial residue such as coffee waste. It contains carbohydrates, proteins, lipids and other micronutrients and can serve as substrate for microbial processes and production of value added products. Solid state fermentation was carried out using coffee cherry husk, coffee pulp, parchment husk, silver skin and spent waste, as substrates for α- amylase production by Neurospora crassa NS1. The parameters were optimized for enzyme synthesis such as moisture level, temperature and pH. The coffee pulp and mixed showed the highest enzyme activity 3908 Units/g and 3870.8 Units/g respectively. The pretreatment of substrate showed increase in 1.8 fold enzyme activity. The supplements like carbon source, nitrogen source and other micronutrients also showed increase in enzyme activity. Overall, Neurospora crassa NS1 shows best α-amylase activity with 7084.4 Units/g by utilizing coffee waste.
